Abstract
Smart drug delivery systems (SDDS) represent a new era in pharmaceutics, combining responsiveness, targeting, and personalization to overcome the limitations of conventional therapies. By harnessing internal and external stimuli, SDDS enable precise, controlled release of drugs while minimizing systemic toxicity. Platforms such as polymeric nanoparticles, liposomes, hydrogels, and bioinspired carriers are being explored for applications in cancer, neurological disorders, metabolic diseases, infectious diseases, and gene therapies. Despite their promise, challenges in biocompatibility, large-scale manufacturing, regulatory approval, and equitable access remain. This review highlights the principles, platforms, applications, global research landscape, and limitations of SDDS, emphasizing their potential to shape precision and personalized medicine for the future.
